Dans cette \u00e9tude, nous proposons de combiner des approches syst\u00e9miques \u00e0 haut d\u00e9bit pour \u00e9tudier l\u2019effet du vieillissement sur la signalisation TCR et la formation de synapses immunologiques dans les cellules T humaines et de souris.<\/p>\n<p><b><i>\u00a0<\/i><\/b><\/p>\n<h2>R\u00f4le des cellules B associ\u00e9es \u00e0 l\u2019\u00e2ge dans l\u2019immunit\u00e9 humorale<\/h2>\n<p><b><i><img loading=\"lazy\" decoding=\"async\" class=\"wp-image-1077 alignleft\" src=\"https:\/\/ihuhealthage.fr\/wp-content\/uploads\/2020\/05\/unnamed-2.png\" alt=\"\" width=\"142\" height=\"71\" \/><\/i><\/b>Ce projet a pour objectif d\u2019\u00e9tudier le r\u00f4le des cellules B associ\u00e9es \u00e0 l\u2019\u00e2ge dans les r\u00e9ponses du centre germinal et la production d\u2019anticorps \u00e0 haute affinit\u00e9.<\/p>\n<h2><b><i>\u00a0<\/i><\/b><\/h2>\n<h2>Caract\u00e9risation fonctionnelle des cellules dendritiques au cours du vieillissement<\/h2>\n<p><b><i><img loading=\"lazy\" decoding=\"async\" class=\"wp-image-1077 alignleft\" src=\"https:\/\/ihuhealthage.fr\/wp-content\/uploads\/2020\/05\/unnamed-2.png\" alt=\"\" width=\"142\" height=\"71\" \/><\/i><\/b>Le d\u00e9veloppement d\u2019une r\u00e9ponse immunitaire est r\u00e9gul\u00e9 par des interactions complexes entre les cellules dendritiques (DC) et les cellules T. Des sous-ensembles distincts de DC sont r\u00e9partis dans tout l\u2019organisme et peuvent \u00eatre distingu\u00e9s en fonction de marqueurs ph\u00e9notypiques, des programmes transcriptionnels, de la distribution et de la fonction des tissus. Notre hypoth\u00e8se est que le vieillissement peut moduler le r\u00e9seau de facteurs sp\u00e9cifiques des tissus qui instruisent la fonction DC pendant la diff\u00e9renciation. Notre projet vise \u00e0 caract\u00e9riser ce r\u00e9seau afin d\u2019identifier de nouvelles cibles pour reprogrammer la fonction DC chez les personnes \u00e2g\u00e9es.<\/p>\n<p><b><i>\u00a0<\/i><\/b><\/p>\n<h2>Infection par le G\u00e9notype 3 du virus de l\u2019h\u00e9patite E chez la personne \u00e2g\u00e9e et r\u00e9ponse immunitaire<\/h2>\n<p><b><i><img loading=\"lazy\" decoding=\"async\" class=\"wp-image-1077 alignleft\" src=\"https:\/\/ihuhealthage.fr\/wp-content\/uploads\/2020\/05\/unnamed-2.png\" alt=\"\" width=\"142\" height=\"71\" \/><\/i><\/b><br \/>\nLe vieillissement est associ\u00e9 \u00e0 une sensibilit\u00e9 accrue \u00e0 l\u2019infection par divers agents pathog\u00e8nes, y compris le g\u00e9notype 3 du virus de l\u2019h\u00e9patite E (HEV-3). Nous \u00e9tudierons dans un mod\u00e8le animal, comment l\u2019infection par un parasite module le cours de la maladie, tant au niveau pathologique que comportemental et nous \u00e9tudierons quelles populations de cellules immunitaires peuvent contribuer \u00e0 la pathogen\u00e8se. Ces \u00e9tudes animales seront compar\u00e9es \u00e0 nos r\u00e9sultats concernant les populations de cellules immunitaires humaines que nous aurons identifi\u00e9es dans le sang de patients \u00ab\u00a0fragiles\u00a0\u00bb, suivis dans la cohorte \u00ab\u00a0COG-FRAIL\u00a0\u00bb.
